MSSQL and PHP bottleneck

I'm currently using PHP in sync with IIS and SQL Server. Right now I'm having a bottleneck issue. I have PHP talking to Server One, which goes to Server Two. Server Two has the database. Server One holds IIS. I thought that could've been an issue. So I moved the database to Server One. There is a noticeable improvement but I'm still getting a page loading slow every... few minutes or so.

Is there some typeime of optimization I could do in SQL Server or in IIS that might fix this issue?

A bit more info, when I had Server One talking to Server Two I'd get a complete timeout. Now, moving the databases to Server One I don't get timeouts but as I said I get a slow loading page every now and then.
